| preparation | dried and milled whole grass (3.0) with acetone (20 L) at room temperature soak 48 h, filtrate decompression recovery solvent, the same method to extract three times, get 92.0 g Acetone Extract. The medicinal material residue was further soaked with methanol (12L) for 72h, and the filtrate was extracted twice under reduced pressure to obtain 222.6 g of methanol extract. The Acetone Extract and methanol extract were combined, dissolved in 90% methanol, and extracted with n-hexane and dichloromethane successively to obtain 71.0g of n-hexane extract, 30.4 g of dichloromethane extract and 197.5g of water fraction. The water fraction (197.5 g) was passed through D101 macroporous resin and eluted sequentially with water, 20%, 40%, 60% and 80% ethanol-water to obtain water-eluting fraction Fr.W1 (about 160 g), 20% ethanol fraction Fr.W2(13.2g), 40% ethanol fraction Fr.W3(15.0g), 60% ethanol fraction Fr.W4 (about 3.0 g) and 80% ethanol fraction Fr.W5 (about 2.0g). Fr.W2 was subjected to Sephadex LH-20 column chromatography eluting with 50% methanol to obtain 8 fractions (Fr. W2a-w2h). Fr.W2d compound l(16.4 mg) was purified by silica gel column chromatography and semi-preparative HPLC. |
